Fast Growth of Eucommia Antifungal Protein(EAFP) Crystals Observed by Atomic Force Microscopy

摘要:The eucommia antifungal protein (briefly named as EAFP) is an antifungal protein extracted from the barkes of the eucommia ulmoides Oliv[1]. It is involved in the defense mechanism of plants and plays an important role against the pathogenic attacks. EAFP has 41 residues distinctly with five disulfide bridges. It has been crystallized in a monoclinic form( P21 ) with the unit cell parameters a = 19. 085, b = 23. 225, c = 30. 854?, β = 98.64[2]. EAFP crystals grow much faster than most of other macromolecular crystals. It takes only several hours to grow perfect crystals big enough for x - ray diffraction at very high resolution( about 0.8?). Therefore it is very interesting to know the mechanism of the fast growth process of EAFP crystals.Here we report step growth rates measured from images by in situ Atomic Force Microscopy(AFM).

  • Experimental Simulation of Platelet Adhesion to a Semi- Permeable Wall Exposed to Flow Disturbance

    Adherent platelets are an important part of both thrombus formation and in certain stages of atherogenesis. Both the formation of atherosclerotic lesions and deposition of platelet thrombi develop not randomly and not everywhere in the circulation, but localize at certain particular areas in the circulation. In fact, platelet thrombi in vivo have been found at sites downstream from bifurcations stenoses and venous valves. In these areas, the blood flow is disturbed and separation of streamlines from the vessel wall and formation of vortices (eddies) are likely to occur. This strongly suggests the involvement of hemodynamic factors in the localization of the vascular diseases.

  • 动脉脱细胞三维胶原支架主要组织相容复合体I(MHC I)抗原的免疫组化观察

    采用异种动物血管制作组织工程血管的支架,再种植宿主自体平滑肌细胞和内皮细胞,是目前较理想的构建组织工程血管的模型.然而,胶原纤维、弹力纤维、细胞外基质和脱细胞后残留的细胞碎片都具有抗原性,尤其是细胞膜上的MHC是一种强烈的移植抗原,如何处理生物支架的抗原性是构建全生物化组织工程血管的一个关键问题之一.同时,在支架种植细胞前检测其抗原性的强弱也是必须的基础技术之一.本文选择血管壁组织细胞高表达的MHC I作为检测指标,观察血管支架处理前后MHC I的变化情况.

  • Using SIMM To Construct Musculoskeletal Modeling of the Human Leg

    Introduction The researches show that a realistic three - dimensional biodynamic model of the human leg is developing into powerful tools for functional analysis of the knee, for evaluation of surgical and diagnostic procedures, and for design of artificial joint[1,2]. In the other word, if it could be established, then it may become possible to accurately obtain the muscle,ligament, and the articular contact forces at the human knee joint in vivo with the aid of biodynamic simulation. This, without doubt, will provide a powerful way for the clinic rehabilitation. But, unfortunately, so far this work is still challenging for biomechanical/biomedical researchers.

  • Development of A Flow Chamber for Studying Cell Behaviour under Pulsatile Flow

    Introduction Because of the complexity and sophistication of the in vivo environment, to systematically study the behavior of cellular response to mechanical forces has mainly relied on the use of in vitro preparations. The parallel plate flow chamber is the most common used for applying fluid shear stress to cultured cells. Most studies used parallel plate flow chambers under steady flow, and only a limited number of in vitro studies concerned with the influence of pulsatile flow. Several kinds of flow chamber were developed which can provide pulsatile flow to cells ( Helmilinger et al , 1991; Ruel et al. , 1995 ;Tacobes et al. , 1998).

  • The Effect of Concentration on Confirmational Transition of Fibroin

    Introduction The silk fibers of silkworm have extraordinary mechanical properties. People commonly recognize that the ?- sheet structure in it mainly contributes that. So the formation of silk fiber from fibroin gel should be studied. Fibroin is primarily synthesized in the posterior division of silk gland of fifth - instar silkworm larva and from the posterior part to the anterior part of silk gland, the concentration of solution increases as well as a conformational transition, ? - helix to ? - sheet transition, occurs simultaneously. But little is known about the detailed process of the conformational transition under the influence of the concentration of fibroin gel. The present work studies the conformational transition of fibroin solution when it is diluted.

  • Experimental Study on the Effects of Femoral Head's Osteonetosis on the Biomechanical Properties of the Femoral Head's Cancellous Bone

    Do the tension、compress and bend test on the normal and morbid femoral head cancellous bone and do the stress relaxation and creep test on the lognitudinal and the transverse samples. Then compare the mechanical natures of the normal and the morbid femoral head's cancellous bone. The object of this research is to offer the basic theory for the research of artificial joint material、the design of artificial joint、the implantation of artificial femoral head and the bone's creation and rebuilding.

  • 声波刺激影响烟草膜蛋白结构和功能的研究

    生物膜是由蛋白质、脂质和碳水化合物等构成的超分子体系,其主要的功能包括能量转换、物质运输和信息传递以及承担着蛋白质和酶系统的代谢进程.质膜是细胞中对环境刺激为敏感的部位,膜蛋白又是膜功能的主要执行者,因此研究声波刺激下膜蛋白结构和功能的改变将有助于了解植物对机械刺激的原初响应过程.本研究以烟草愈伤组织为材料,来研究声波刺激对膜蛋白二级的影响.

  • 神经元自发放电的慢波振荡形式及其通道探讨

    本研究从损伤DRG传人纤维中引导单根神经纤维放电,采集神经纤维放电的动作电位间期(interspike interval,ISI)序列.观察到一种周期为50s~200s,波幅为15ms~50ms的慢波振荡节律形式,该节律往往可以由K+通道阻断剂TEA诱发产生.

  • 肿瘤抑制因子PTEN在高血压动脉重建中的作用初探

    肿瘤抑制因子PTEN(Phosphatase and TENsion homology deleted on chromosome 10)是新近发现的具有磷脂磷酸酶和蛋白磷酸酶双重活性的磷酸酶.人体的多种肿瘤与PTEN基因突变有关.PTEN参与多种细胞增生、迁移、粘附、凋亡、细胞周期等功能的调控.然而,PTEN对心血管系统作用的研究却甚少,它是否参与了应力介导的动脉重建尚未见报道.本文旨在观察血管紧张素转换酶抑制剂卡普托利对高血压大鼠主动脉PTEN mRNA表达的影响,探讨PTEN是否参与了高血压动脉重建.

  • 盘基网柄菌(Dictyostelium discoideum)聚集模式的一种离散模型

    盘基网柄菌(Dictyostelium discoideum)因为其独特的生命周期成为发育生物学特别是生物模式形成广泛研究的模式生物.网柄菌的单倍体细胞从孢子壳中孵化,像陆地阿米巴一样生活,并通过细胞有丝分裂无性生殖.当食物供给耗尽时,阿米巴会在某个区域聚集,并在其点上集合成蛞蝓(slug)状聚集体.而当此蛞蝓迁移到适当的地方,又将形成植物状的子实体并释放新的孢子细胞.网柄菌的生命周期对于多细胞生命体来说虽然是不典型的,但是它的一些发育特征却可作为高等真核生物中相似事件如聚集,细胞分化,模式形成等的事例进行研究.

  • 网状血管计算机模拟分析

    多级分叉或网状血管的流量计算历来是临床和基础研究中受到关注的问题,如局部血管发生关闭或栓塞后对周围血流的影响,产生侧支循环或实施旁路搭接手术后下游血流灌注状况的改善,血管内注入药物后血流分配情况等等.传统的计算方法有水管网络迭代法和电路网络模拟,这些方法在处理边界条件和非线性参数方面存在不够直接和灵活的缺点,本文采用有限元技术和图形处理相结合的方法建立计算机仿真模型分析血管系统中的血流分布.

  • 微管在细胞可变形性中的作用--抗拒微丝产生的张力

    细胞的形态和力学性质或者说细胞的可变形性,决定了细胞对载荷的敏感程度.例如,剪应力持久作用可导致血管内皮细胞长轴沿流场方向排列,同时伴随细胞弹性模量的显著增高以及作用在细胞表面的剪应力梯度的显著降低.与此相对应,同一剪应力条件不再能诱导重排后的血管内皮细胞产生相应的生物学效应,如细胞膜上牵拉敏感离子通道的开放、胞内Ca2+浓度和血小板衍生生长因子(PDGF)B链mRNA的转录水平的增高等.这些证据表明,细胞的可变形性是细胞得以分辨其已经适应的和未适应的载荷的一种机制.

  • 血管壁外膜对张开角的影响

    张开角是指血管壁的圆环试样沿径向剪开时,圆环的周向张开角度.张开角被认为是血管壁残余应变的度量.血管壁应力分布取决于残余应力的大小.李晓阳提出,若沿径向将血管壁分为内外两层,则两层的零应力状态不同,表现为内外两层的张开角不同,内存张开角小于外层.本文实验测量犬主动脉的张开角,讨论血管外膜对张开角的影响和内外层血管张开角的差别.(图1和图2)

  • Stretching Alpha Helix of 18 Peptide by Computer Simulation

    Aims Molecular biomechanics is emerging as a new field in recent years [1]. But the experiment methods now limited by the specimen preparing, which relies on the complicate physical and chemical operation of single molecule. On computer by software, it is much more convenient to do such stretching by simulation. And Insight Ⅱ [5] software, running on Irix system,provides the convenience for us in this field.

  • 组织工程化肌腱修复鸡趾屈肌腱缺损的生物力学特性研究

    目的肌腱缺损的修复与功能重建是骨外科的研究重点之一.组织工程化肌腱为修复肌腱缺损开辟了一条新途径.它具有良好的生物相容性、生物活性、生物力学相容性并能诱导胶原生成.近年来,在组织工程化肌腱方面已开展了多方面的研究,初步临床应用也取得了较好的效果.但是,从组织工程化肌腱的研究和应用中发现,它在植入体内后,其抗拉强度始终达不到正常肌腱的数值.不难推断,材料在体内的降解速率及诱导生成的胶原纤维的数量和质量同新生肌腱的生物力学性能有密切关系.

  • 玫瑰花瓣实验的概率动力学模拟

    玫瑰花瓣实验作为考察细胞间特异性相互作用的简单实验,已被免疫学家广泛用于评价受体与配体间的功能性作用,以及区分白细胞亚型[1~6].早期实验是将人T淋巴细胞和羊红细胞混合而发生粘附现象,大的淋巴细胞表面会吸附上小的红细胞,形成玫瑰花瓣结构.由于该实验不能把玫瑰花瓣尺寸分布与分子性质定量地联系起来,故迄今只是一种定性方法.我们根据已建立的小系统概率动力学模型设计新的玫瑰花瓣实验,能够把玫瑰花瓣尺寸分布与受体/配体密度及它们的反应亲和性联系起来,从而为临床免疫检测提供定量依据和方法学基础.
